Abstract:To explore the possible molecular mechanism of Guben Zhibeng Decoction in the treatment of dysfunctional uterine bleeding by means of network pharmacology.Methods:The active components of Guben Zhibeng Decoction were screened on the platform of pharmacology database of traditional Chinese medicine system and their potential action targets were predicted,and then the obtained targets were imported into UNIPROT database for gene standardization treatment; the related targets of dysfunctional uterine bleeding was searched in GeneCards and OMIM databases.The network diagram of “drug-component-target-disease” was constructed by using Cytoscape 3.7.2 software,and the interaction between proteins was analyzed by using STRING online platform.GO biological function and KEGG pathway enrichment were analyzed by DAVID database,and the results were displayed in graph form.Results:A total of 58 effective active ingredients in Guben Zhibeng Decoction were obtained by OB and DL screening criteria,and 135 targets were predicted.A total of 2 164 targets of dysfunctional uterine bleeding were found in GeneCards and OMIM,and 85 targets of intersection of drugs and diseases were found in Venn diagram.The key targets involved IL6,VEGFA,TP53,TNF,EGF,etc.A total of 135 GO items were obtained by P enrichment analysis,including 38 biological processes,36 molecular functions and 61 cellular components,mainly involving in hypoxia response,positive regulation of nitric oxide biosynthetic process,extracellular space,extracellular region,enzyme binding,steroid binding,etc.A total of 25 pathways were found by KEGG enrichment analysis(P<0.05,FDR<0.05),and the target genes were mainly enriched in cancer signal pathway etc.Conclusion:Guben Zhibeng Decoction has the characteristics of multi-component,multi-target and multi-channel in the treatment of dysfunctional uterine bleeding,and its mechanism is complex,which provides material basis and theoretical basis for subsequent experimental verification.
