      目的:研究三子养亲汤治疗过敏性哮喘的作用机制。方法:使用GEO2R找到GSE40889芯片的差异基因,利用GCBI数据库制作差异基因热图,借助DAVID6.8对差异基因进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)富集分析,使用Cytoscape构建蛋白质-蛋白质相互作用网络,筛选核心基因,分析网络,再进一步利用Cytoscape的MCODE插件对差异表达基因进行模块分析。结果:共查询有效成分21个,对应靶点268个;筛选出213个差异基因,差异表达基因主要富集在膜组织、RNA结合及转录翻译方面等,涉及通过TNF-α、B细胞受体、TGF-β、PI3K/AKT信号通路。得到一个重要模块,功能主要集中在基因沉默的调控、蛋白质异构化、细胞-细胞黏附、病毒过程等方面及RNA聚合酶Ⅱ启动子转录的正调控、p53介导的内源性凋亡信号通路对DNA损伤的响应、DNA模板、宿主对病毒转录的正调控等,涉及Notch信号通路、FoxO信号通路、PI3K-AKT信号通路等信号通路。结论:三子养亲汤可能通过TNF-α、B细胞受体、TGF-β、PI3K/AKT信号通路,调控基因沉默、使蛋白质异构化,影响转录翻译等途径发挥作用。
English Summary:
      To study the mechanism of Sanzi Yangqin Decoction in the treatment of allergic asthma.Methods:Gene Expression Omnibus 2R(GEO2R) was used to find the differential gene of GSE40889 chip,and Gene-Cloud Biotechnology Information(GCBI) database was used to make the differential gene heat map.DAVID 6.8 was used to conduct Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analyses on the differential gene.Cytoscape was used to construct the network of protein-protein interaction.The core gene was screened out,and the network was analyzed.The module analysis of the differential gene was carried out by the Molecular Complex Detection(MCODE) of Cytoscape.Results:A total of 21 active components were queried,268 corresponding targets were found.A total of 213 differential genes were screened out,which were mainly enriched in membrane tissue,ribonucleic acid(RNA) binding,transcription,and translation,involving signaling pathways of tumor necrosis factor-α(TNF-α),B cell receptor,transforming growth factor-β(TGF-β),and phosphoinositide 3 kinase/protein kinase B(PI3K/AKT).An important module was obtained,which mainly focused on the regulation of gene silencing,protein isomerization,cell-cell adhesion,virus process,the positive regulation of RNA polymerase II promoter transcription,the response of endogenous apoptosis signaling pathway mediated by tumor suppressor p53 to deoxyribonucleic acid(DNA) damage,DNA template,and the positive regulation of host to virus transcription,involving signaling pathways of Notch,forkhead box O(FoxO),and PI3K-AKT.Conclusion:Sanzi Yangqin decoction may regulate gene silencing,make protein isomerization,and affect transcription and translation through the signaling pathways of TNF-α,B cell receptor,TGF-β,and PI3K/AKT.
