    目的:研究粉防己碱对弗氏完全佐剂(FCA)致慢性炎性疼痛小鼠模型的镇痛作用。方法:建立FCA足底慢性炎性疼痛模型,使用粉防己碱进行干预治疗,结合机械刺激、热辐射刺激、热板实验及伤害性反应等疼痛行为学检测方法鉴定其镇痛作用特点,使用实时PCR、蛋白质印迹法等检测辣椒素瞬时受体电位1(TRPV1) mRNA和蛋白表达变化,探究其干预作用TRPV1敏化调控途径机制。结果:获得了慢性炎性疼痛的小鼠模型。粉防己碱可显著增加该慢性炎性疼痛模型的机械刺激和热辐射刺激痛阈值,却不影响该模型的热板刺激痛阈值及小鼠体质量,与正常对照组比较,粉防己碱干预后,FCA致慢性炎性疼痛小鼠模型的TRPV1产生明显减少(P<0.05)。结论:粉防己碱能显著抑制FCA致慢性炎性疼痛小鼠模型的TRPV1产生,对慢性炎性疼痛具有镇痛作用,且未发现耐受性及毒性。

    Abstract:

    To investigate the analgesic effect of tetrandrine(TEF) on chronic inflammatory pain induced by Freund's complete adjuvant(FCA) in mice.Methods:A FCA plantar chronic inflammatory pain model was established,and TEF was used to conduct intervention treatment.The analgesic effect of TEF was determined by pain behavioral tests of mechanical stimulation,thermal radiation stimulation,hot-plate test,and nociceptive response.Real-time quantitative polymerase chain reaction(real-time PCR),Western blot and other methods were used to detect the messenger ribonucleic acid(mRNA) and protein expression changes of transient receptor potential vanilloid-1(TRPV1),and to explore the regulatory pathway and mechanism of TRPV1 sensitization.Results:The model of chronic inflammatory pain in mice was obtained.TEF significantly increased the pain thresholds of mechanical stimulation and thermal radiation stimulation in mice,but did not affect the pain threshold of hot plate stimulation and the weight of mice.Compared with the control group,TRPV1 in the mice with FCA-induced chronic inflammatory pain was significantly reduced after the intervention of TEF(P<0.05).Conclusion:TEF significantly inhibited the production of TRPV1 in the mice with FCA-induced chronic inflammatory pain,and had analgesic effect on chronic inflammatory pain.No tolerance or toxicity of TEF has been found.
