目的:研究小檗碱调控核因子κB信号转导通路对幽门螺杆菌(Hp)感染人胃癌细胞的影响。方法:建立Hp感染人胃癌细胞模型,并将其分成对照组、黄连素组、5-氟尿嘧啶组以及黄连素+5-氟尿嘧啶组。其中,对照组则予以等量生理盐水干预;黄连素组予以100 μg/mL浓度小檗碱干预;5-氟尿嘧啶组予以5 μg/mL的5-氟尿嘧啶干预;黄连素+5-氟尿嘧啶组则同时予以上述剂量的小檗碱以及5-氟尿嘧啶干预。比较各组Hp感染人胃癌细胞增殖情况,细胞周期的改变,核因子κB信号通路相关蛋白表达,炎症反应指标水平。结果:对照组、黄连素组、5-氟尿嘧啶组以及黄连素+5-氟尿嘧啶组干预后2 d、3 d时的胃癌细胞增殖率呈逐渐下降趋势;对照组、黄连素组、5-氟尿嘧啶组以及黄连素+5-氟尿嘧啶组感染人胃癌细胞G0/G1期、S期呈逐渐下降趋势,而G2/M期呈逐渐升高趋势;对照组、黄连素组、5-氟尿嘧啶组以及黄连素+5-氟尿嘧啶组IκB激酶α蛋白表达水平呈逐渐升高趋势,而P65蛋白表达水平呈逐渐降低趋势;对照组、黄连素组、5-氟尿嘧啶组以及黄连素+5-氟尿嘧啶组IL-4水平呈逐渐升高趋势,而白细胞介素-6呈逐渐下降趋势,各组比较差异均有统计学意义(均P<0.05)。结论:小檗碱主要是通过调控核因子κB信号转导通路相关蛋白的表达,继而发挥抑制Hp感染人胃癌细胞增殖与炎症反应的作用,值得临床重点关注。 |

To investigate the effect of berberine regulation of the nuclear factor κB(NF-κB) signaling pathway on human gastric cancer cells infected with Helicobacter pylori(Hp).Methods:The Hp-infected human gastric cancer cell model was established and the cells were divided into a control group(normal saline),a berberine group(100 μg/mL),a 5-fluorouracil(5-Fu) group(5 μg/mL),and a combination group(100 μg/mL berberine+5 μg/mL 5-Fu).The proliferation,cell cycles,protein expression related to the NF-κB signaling pathway,and inflammatory response index levels of Hp-infected human gastric cancer cells in the four groups were compared.Results:The proliferation rates of gastric cancer cells in all groups showed a decreasing trend on the 2nd day and 3rd day of the intervention.The Hp-infected human gastric cancer cells in the G0/G1 and S phases in all groups showed a gradually decreasing trend,while those in the G2/M phase showed a gradually increasing trend.The expression level of IκBα protein in all groups showed a gradually increasing trend,while the expression level of P65 protein showed a gradually decreasing trend.The level of IL-4 in all groups showed a gradually increasing trend,while the level of IL-6 showed a gradually decreasing trend.There were significant differences among all groups(all P<0.05).Conclusion:Berberine inhibits the proliferation and inflammatory response of Hp-infected human gastric cancer cells by regulating the expression of proteins related to the NF-κB signaling pathway,which is worthy of clinical attention. |
