      目的:探讨温和灸足三里穴对自然衰老大鼠的肾保护作用及其机制。方法:雄性SD大鼠随机数字表法分为自然衰老组和艾灸足三里组,同时设立9个月龄对照组,每组10只。自然衰老组常规饲养至20个月龄,艾灸足三里组从14个月龄开始温和灸“足三里”穴,1次/d,20 min/次,两侧穴位交替进行,直至20个月龄。结果:与9个月龄对照组比较,自然衰老组大鼠毛发枯黄,精神不振,活动欠灵活;肾结构改变,肾间质出现炎细胞浸润以及大量蓝色胶原纤维,肾组织中β-半乳糖苷酶表达升高(P<0.05),24 h尿蛋白含量升高(P<0.05),大鼠肾组织纤连蛋白(FN)、转化生长因子-β1(TGF-β1)mRNA表达均增加(P<0.01)。与自然衰老组比较,艾灸足三里组大鼠毛发润泽,色泽微黄,活动灵活,日常活动均有不同程度改善;胶原染色减少,肾组织中β-半乳糖苷酶表达降低(P<0.05),24 h尿蛋白含量降低(P<0.05),肾组织中FN、TGF-β1 mRNA表达降低(P<0.05)。结论:温和灸足三里穴能减轻大鼠因增龄造成的肾损伤,对自然衰老大鼠具有肾脏保护作用,可能与抑制肾组织FN表达及TGF-β1基因转录有关。
English Summary:
      To explore the renal protective effect and mechanism of mild moxibustion at Zusanli on naturally aging rats.Methods:Male SD rats were randomly divided into a natural aging group(n=10) and a moxibustion group(n=10) according to a random number table.Ten 9-month-old rats were assigned to the control group.The rats in the natural aging group were fed to the age of 20 months routinely.The rats in the moxibustion group underwent moxibustion at Zusanli from the age of 14 months,once a day for 20 min each time.The acupoints on both sides were selected alternately until the rats aged 20 months.Results:Compared with the control group,the natural aging group showed dry and yellow fur,low energy,stiff movement,altered renal structure,inflammatory cell infiltration and massive blue collagenous fibers in the renal interstitium,increased expression of β-galactosidase in renal tissues(P<0.05) and the content of 24-hour urinary protein(P<0.05),and elevated mRNA expression of fibronectin(FN) and transforming growth factor(TGF)-β1 in renal tissues(P<0.01).Compared with the natural aging group,the moxibustion group showed glossy and yellowish fur,flexible and improved daily activity,reduced collagen staining,reduced expression of β-galactosidase(P<0.05) and the content of 24-hour urinary protein(P<0.05) in renal tissues,and declining mRNA expression of FN and TGF-β1 in renal tissues(P<0.05).Conclusion:Mild moxibustion at Zusanli can alleviate renal injury induced by aging,and has a protective effect on the kidney in naturally aging rats.The mechanism may be related to the inhibition of FN protein expression and TGF-β1 gene transcription in renal tissues.
