Abstract:To explore the efficacy and potential targets of Fangji Huangqi Decoction against chronic heart failure.Methods:Immunofluorescence and reverse transcription-quantitative polymerase chain reaction (RT-qPCR) were used to determine the expression of atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP),the markers of phenylephrine-induced myocardial hypertrophy and chronic heart failure,in H9c2 cells.The active components and targets of Fangji Huangqi Decoction were retrieved from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and previous research,and the main targets of chronic heart failure from Genecards and other databases.Thereby,the common targets of the disease and the decoction were screened out.The interaction between the targets was analyzed based on STRING and the protein-protein interaction (PPI) network was constructed.DAVID and ClusterProfiler were employed for Gene Ontology (GO) term and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment.Cytoscape 3.7.2 software was used to plot the networks.Results:Fangji Huangqi Decoction could alleviate the phenylephrine-induced myocardial hypertrophy and decrease the expression of ANP and BNP genes.The 166 anti-chronic heart failure targets of Fangji Huangqi Decoction were involved in 493 biological processes and 171 signaling pathways.Conclusion:Fangji Huangqi Decoction has therapeutic effect on chronic heart failure in vitro,mainly.To be specific,the active components quercetin,formononetin,tetrandrine,and astragaloside IV acts on hypoxia-,inflammation-,and metabolism-related signaling pathways,to prevent and treat chronic heart failure.
