Abstract:To optimize the extraction process of total flavonoids from Psychotria asiatica L.using response surface methodology followed by evaluation with enrichment technology by macroporous adsorption resin.Methods:On the basis of single factor experiment,with the ethanol concentration,extraction temperature,solid-liquid ratio,and extraction time as independent variables and the yield of total flavonoids as the response value,the process conditions were optimized by the Box-Behnken experiment.Moreover,the optimum macroporous resin was screened out in the enrichment process using adsorption,desorption,and recovery effect as indexes.Results:The results showed that the optimal extraction conditions were as follows:extraction temperature of 59 ℃,ethanol concentration of 75%,solid-liquid ratio of 1:10,extraction time of 48 min,and extraction twice.Under the optimized conditions,the extraction rate was 25.60% and the yield of total flavonoids was 2.36%.The HP-20 resin showed a good enrichment effect on total flavonoids of P.asiatica.After enrichment,the recovery rate of total flavonoids reached 87.70%,and the purity of total flavonoids was 6 times higher than that of the crude extract.Conclusion:The optimized process is simple and stable and can provide references for the development and utilization of P.asiatica L.
