目的:观察滋肾醒脑汤对阿尔茨海默病(AD)大鼠的学习记忆能力及海马组织Caspase-3、Caspase-9的表达影响。方法:采用侧脑室注入β淀粉样蛋白25-35制备AD大鼠模型,同时采用滋肾健脑汤进行干预,采用Morris水迷宫检测滋肾醒脑汤对AD模型大鼠学习记忆能力及空间探索能力的影响,通过免疫组织化学法观察大鼠海马CA1区Caspase-3、Caspase-9的表达。结果:Morris水迷宫试验:模型组大鼠学习记忆能力较假手术组明显下降(P<0.05)。苏木精-伊红(HE)染色结果显示:模型组大鼠海马神经元细胞损伤严重,而中药组和阳性对照组神经元细胞损伤情况均较模型组减轻。免疫组织化学结果:模型组大鼠海马区Caspase-3、Caspase-9明显增加(均P<0.05),而中药组能明显降低Caspase-3、Caspase-9的表达(均P<0.05)。结论:滋肾醒脑汤可减轻大鼠海马神经元的损伤,提高AD大鼠学习记忆能力,其机制可能与抑制海马组织中Caspase-3、Caspase-9的表达有关。 |

To observe the effect of Zishen Xingnao Decoction on the learning and memory ability of Alzheimer's disease (AD) rats and the expression of Caspase-3 and Caspase-9 in hippocampus.Methods:In this experiment,β-amyloid (Aβ25-35) was injected into the lateral ventricle to induce AD in rats and the Zishen Jiannao Decoction was used for intervention.Morris water maze was employed to test the effect of Zishen Xingnao Decoction on the learning and memory ability and space exploration ability of AD rats.The expression of Caspase-3 and Caspase-9 in the CA1 area of rat hippocampus was determined with the immunohistochemical method.Results:In the Morris water maze test,the model group showed lower learning and memory ability than the sham operation group (P<0.05).According to the hematoxylin and eosin (HE) staining result,the model group had severe injury of neuronal cells in the hippocampus,while the injury was milder in the Chinese medicine group and the positive control group than in the model group.The expression of Caspase-3 and Caspase-9 in the hippocampus of the model group increased (P<0.05) and that in the Chinese medicine group decreased (P<0.05).Conclusion:Zishen Xingnao Decoction can alleviate the damage of hippocampal neurons and improve the learning and memory ability of AD rats.The mechanism is the likelihood that it inhibits the expression of Caspase-3 and Caspase-9 in the hippocampus. |
