目的:运用网络药理学方法探究丹参饮治疗心绞痛的作用机制。方法:本研究通过中药系统药理学数据库与分析平台(TCMSP)筛选出丹参饮中丹参、檀香、砂仁的有效活性成分及其相关靶点蛋白,通过GeneCards数据库挖掘心绞痛的疾病靶点,将其与药物靶点蛋白取其交集,并将交集靶点运用Cytoscape 372构建活性成分-靶点-疾病网络图。运用STRING数据库对交集靶点构建蛋白质-蛋白质相互作用(PPI)I网络图,并进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)通路富集分析。结果:共筛选出丹参饮的活性成分78个,靶点334个,心绞痛的靶点2 578个,其交集靶点93个,对应的活性成分59个。GO生物过程20条,包括肾上腺素能受体活性、G蛋白偶联胺受体活性、泛素样蛋白连接酶结合、细胞因子受体结合、酰胺结合等;KEGG相关信号通路,涉及HIF-1信号通路、IL-17信号通路、T细胞受体信号通路等。结论:本研究体现了丹参饮治疗心绞痛的作用机制,体现了中药多成分、多靶点、多途径的作用特点,为临床应用及药理学研究提供了思路和依据。 |

To explore the mechanism of Danshen Decoction in the treatment of angina pectoris by means of network pharmacology.Methods:The active components and corresponding targets of Salviae Miltiorrhizae Radix et Rhizoma,Santali Albi Lignum,and Amomi Fructus in Danshen Decoction were screened from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the targets of angina pectoris from GeneCards.The common targets shared by Danshen Decoction and angina pectoris were intersected to construct the active component-target-disease network in Cytoscape 372.The protein-protein interaction(PPI) network of the common targets was constructed by STRING,and the gene ontology(GO) functional annotation and Kyoto Encylopedia of Genes and Genomes(KEGG) pathway enrichment analysis were carried out for the targets.Results:A total of 78 active components and 334 targets of Danshen Decoction and 2 578 targets of angina pectoris were mined.Danshen Decoction and angina pectoris shared 93 common targets which corresponded to 59 active components.The common targets were involved in 20 GO biological processes,including adrenergic receptor activity,G protein-coupled amine receptor activity,ubiquitin-like protein ligase binding,cytokine receptor binding,and amide binding.The enriched KEGG signaling pathways included HIF-1,IL-17,and T cell receptor signaling pathways.Conclusion:This study predicts the mechanism of Danshen Decoction in the treatment of angina pectoris,which involves multiple components,targets and pathways.The results provide a basis for relevant clinical application and pharmacological research. |
