    目的:通过构建药物与疾病之间的网络关系探究黄芪对缺血性中风(IS)的治疗机制。方法:在中药系统药理学数据库与分析平台(TCMSP)检索黄芪有效活性成分和相关靶点;以“Cerebral Ischemic Stroke”为关键词,在人类基因的综合数据库(GeneCards)和在线人类孟德尔遗传数据库(OMIM)检索IS疾病基因靶点;利用Perl脚本分析计算出黄芪-IS之间的共同基因,String网站用来构建黄芪-IS之间蛋白作用关系网络,黄芪-IS交集基因的GO功能富集分析、KEGG通路富集分析则借助R软件完成。结果:从黄芪中筛选出16个有效成分,作用于137个IS疾病靶点,黄芪治疗IS的核心基因主要有AKT1、IL6、VEGFA、CASP3、JUN等。中药-疾病靶点共有144条GO生物学过程,功能主要与细胞活性、特异性DNA的表达和酶反应有关。共获得164条KEGG信号通路,主要包括AGE-RAGE信号通路、乙型肝炎病毒通路、前列腺癌通路、TNF信号通路、人巨细胞病毒感染通路、膀胱癌通路、胰腺癌通路、丙型肝炎通路、内分泌抵抗通路等。结论:黄芪可通过多成分、多靶点和多通路治疗IS,为临床用药提供科学理论支撑。

    Abstract:

    To explore the therapeutic mechanism of Astragali Radix on ischemic stroke(IS) by constructing network relationship between the drug and the disease.Methods:The active components and related targets of Astragali Radix were retrieved in the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The gene targets of IS were retrieved from GeneCards and Online Mendelian Inheritance in Man(OMIM) with the term “cerebral ischemic stroke”.Perl script was used to analyze and calculate the common genes of Astragali Radix and IS.String was used to construct the protein-protein interaction(PPI) network of Astragali Radix and IS.The GO function enrichment analysis and KEGG pathway enrichment analysis of common genes of Astragali Radix and IS were carried out by R software.Results:Sixteen active components of Astragali Radix were screened out,which acted on 137 IS-related targets.The core genes of Astragali Radix in the treatment of IS mainly included AKT1,IL6,VEGFA,CASP3,and JUN.The targets of the drug and the disease involved 144 GO biological processes,and their functions were mainly related to cell activity,specific DNA expression,and enzyme reactions.A total of 164 KEGG signaling pathways were obtained,including AGE-RAGE signaling pathway,hepatitis B virus pathway,prostate cancer pathway,TNF signaling pathway,human cytomegalovirus infection pathway,bladder cancer pathway,pancreatic cancer pathway,hepatitis C pathway,and endocrine resistance pathway.Conclusion:Astragali Radix can treat IS through multiple components,multiple targets,and multiple pathways,which provides certain scientific theoretical support for Astragali Radix in the treatment of IS.
