目的:分离纯化铁皮石斛多糖(DOP),得到纯化后的铁皮石斛多糖DOP-1,并以秀丽隐杆线虫为模式生物,研究DOP-1的抗衰老作用。方法:以铁皮石斛鲜品为原料,经提取、浓缩、乙醇沉淀和真空干燥后,得到铁皮石斛粗多糖,铁皮石斛粗多糖经DEAE-Sepharose Fast Flow层析柱和HW-55F凝胶柱洗脱后获得纯品铁皮石斛多糖DOP-1,通过高效凝胶渗透色谱法测定其纯度及相对分子质量等参数,同时研究不同浓度DOP-1(100 μg/mL、200 μg/mL、400 μg/mL)对秀丽隐杆线虫寿命、生殖能力、抗高温损伤、氧化应激损能力及脂褐素的影响。结果:分离纯化后的DOP-1为均一性多糖,其相对分子质量为3.260×105,不同浓度DOP-1均可以显著延长秀丽隐杆线虫的寿命(P<0.05或P<0.01),其中以200 μg/mL DOP-1作用最为明显,与对照组比较,寿命延长了43.37%,DOP-1对高温损伤和氧化应激损伤有一定的抵御作用,能有效降低秀丽隐杆线虫体内脂褐素的积累,并对其生殖能力无明显影响。结论:分离纯化后的DOP-1为均一性多糖,并具有较显著的抗衰老作用,可对其结构与抗衰老活性之间关系的研究提供参考。 |

To separate and purify Caulis Dendrobii Officinalis polysaccharides(DOP) to obtain DOP-1 and to study the anti-aging effect of DOP-1 with Caenorhabditis elegans.Methods:Fresh Caulis Dendrobii Officinalis was extracted and the extract was concentrated,followed by ethanol precipitation and vacuum drying to yield crude DOP.Crude DOP was separated on DEAE-Sepharose Fast Flow column and eluted with HW-55F gel column to result in the pure DOP-1.Parameters such as purity and relative molecular weight were determined by high-performance gel-permeation chromatography.Moreover,the effect of different concentration of DOP-1(100 μg/mL,200 μg/mL,400 μg/mL) on lifespan,reproductive ability,and lipofuscin accumulation of C.elegans,and the resistance of C.elegans to high temperature and oxidative stress was investigated.Results:The isolated and purified DOP-1 was a homogeneous polysaccharide with relative molecular weight of 3.260 × 105 Da.Different concentration of DOP-1 could significantly prolong the lifespan of C.elegans(P<0.05 or P<0.01),particularly the 200 μg/mL DOP-1 which increased the lifespan by 43.37% compared with the control group.DOP-1 can protect C.elegans from high temperature injury and oxidative stress injury and effectively reduce the accumulation of lipofuscins.However,it has no remarkable influence on the reproductive ability of the C.elegans.Conclusion:The isolated and purified DOP-1 is a homogenous polysaccharide with significant anti-aging effect,which can serve as a reference for the research on the relationship between its structure and anti-aging activity. |
