目的:研究下瘀血汤对肝癌细胞的作用以及对Nanog信号通路的影响。方法:将HepG2细胞分为空白血清组、低剂量含药血清组、中剂量含药血清组;高剂量含药血清组,通过MTT、流式细胞仪、Transwell测肝癌细胞增殖、凋亡及侵袭情况,实时PCR、蛋白质印迹法测Nanog mRNA、Bcl-2、胱天蛋白酶-3水平。结果:空白血清组肝癌细胞的OD值及侵袭数量高于中剂量、高剂量含药血清组(P<0.05),肝癌细胞凋亡率低于低剂量、中剂量、高剂量含药血清组(P<0.05),高剂量含药血清组肝癌细胞OD值及侵袭数量最少、凋亡最高;空白血清组肝癌细胞中Nanog mRNA及Bcl-2蛋白表达高于低剂量、中剂量、高剂量含药血清组(P<0.05),胱天蛋白酶-3的蛋白表达水平低于低剂量、中剂量、高剂量含药血清组(P<0.05),与低剂量、中剂量剂量含药血清组比较,高剂量含药血清组肝癌细胞中Nanog mRNA及Bcl-2蛋白表达明显降低(P<0.05),胱天蛋白酶-3蛋白表达显著升高(P<0.05)。结论:下瘀血汤通可抑制肝癌细胞的活性及其侵袭力、促进癌细胞的凋亡,其作用机制可能调控与Nanog信号通路,从而促进胱天蛋白酶-3的表达、抑制Bcl-2水平有关。 |

To study the effect of Xianyuxue Decoction on hepatocarcinoma cells and Nanog signaling pathway.Methods:HepG2 cells were divided into four groups,namely the blank serum group,the low-dose drug-containing serum group,the medium-dose drug-containing serum group,and the high-dose drug-containing serum group.The proliferation,apoptosis,and invasion of HCC cells were measured by methyl thiazolyl tetrazolium(MTT) assay,depletion cell analyzer,and Transwell.The levels of Nanog mRNA,B-cell lymphoma-2(Bcl-2),and caspase-3 were determined by real-time polymerase chain reaction(RT-PCR) and Western blot.Results:OD value and invasion number of hepatocellular carcinoma cells in the blank serum group were higher than those in the medium and high-dose drug-containing serum groups(P<0.05),and apoptosis rate of hepatocellular carcinoma cells was lower than those in the low,medium and high-dose drug-containing serum groups(P<0.05).OD value and invasion number of hepatocellular carcinoma cells in the high-dose drug-containing serum group were the least,and apoptosis was the highest.The expressions of Nanog mRNA and Bcl-2 protein in the blank serum group were higher than those in the low,medium,and high-dose drug-containing serum groups(P<0.05),and the expression of caspase-3 protein in the blank serum group was lower than that in the low,medium,and high-dose drug-containing serum groups(P<0.05).The expression of Nanog mRNA and Bcl-2 protein was significantly decreased in the high-dose drug-containing group(P<0.05) and the expression of caspase-3 protein was significantly increased(P<0.05) as compared with the low and medium-dose drug-containing serum groups.Conclusion:Xiayuxue Decoction can inhibit the activity and invasion ability of hepatocarcinoma cells and promote the apoptosis of cancer cells,and its mechanism may be related to the regulation of Nanog signal pathway,thus promoting the expression of caspase-3 and inhibiting the level of Bcl-2. |
