To study the protective effect of Bushen Jiedu Decoction against placenta injury and oxidative stress in pregnant mice after radiation.Methods:Fifty female mice and 25 male mice were selected.After successful conception,female mice were divided into a blank control group,a low-dose radiation group,a medium-dose radiation group,a low-dose radiation+Bushen Jiedu Decoction group,and a medium-dose radiation+Bushen Jiedu Decoction group according to the random number table,with 10 mice in each group.Mice in the low-and medium-dose radiation+Bushen Jiedu Decoction groups received Bushen Jiedu Decoction at 13.5 g/(kg·d) for intervention,while those in the blank control group,and low-and medium-dose radiation groups were given the equal volume of normal saline by gavage.After 10 days of intervention by gavage,mice,except for those in the blank control group,underwent γ ray radiation.Specifically,the radiation dose in the low-dose radiation group and the low-dose radiation+Bushen Jiedu Decoction group was 2 Gy,and that in the medium-dose radiation group and the medium-dose radiation+Bushen Jiedu Decoction group was 4 Gy.The pregnancy outcome,general condition of neonatal mice,placental index,and oxidative stress indicators of pregnant mice of the two groups were compared.Results:In the low-and medium-dose radiation groups,the live birth rate was lower and the stillbirth rate was higher than those in the blank control group(P<0.05).In the low-and medium-dose radiation+Bushen Jiedu Decoction groups,the live birth rate was higher and the stillbirth rate was lower than those in the low-and medium-dose radiation groups(P<0.05).Compared with the blank control group,the low-and medium-dose radiation groups showed decreased body mass,body length,and tail length of neonatal mice,and placental index(P<0.05).Compared with the low-and medium-dose radiation groups,the low-and medium-dose radiation+Bushen Jiedu Decoction groups showed increased body mass,body length,and tail length of neonatal mice,and placental index(P<0.05).SOD and GSH-Px levels of pregnant mice in the low-and medium-dose radiation groups were lower than those in the blank control group,while MDA levels were higher(P<0.05).The levels of SOD and GSH-Px in the low-and medium-dose radiation+Bushen Jiedu Decoction groups were higher than those in the blank control group,and the levels of MDA were lower(P<0.05).Conclusion:Bushen Jiedu Decoction has a significant protective effect on the placenta injury of pregnant rats after radiation,which is beneficial to reduce oxidative stress injury and improve the pregnancy outcome of pregnant mice.
