      目的:比较结直肠癌血瘀证肿瘤组织与正常组织的差异表达蛋白。方法:采用临床调查方法,对结直肠癌患者进行证候分析,留取血瘀证患者术后标本的肿瘤组织和远端正常肠黏膜组织,运用iTRAQ定量蛋白质组学比较2种组织的差异表达蛋白,并应用Metascape进行富集和分析。结果:在血瘀证组织比较中,肿瘤组织较远端正常组织差异表达蛋白总数为55个,其中上调13个,功能与调节型胞吐作用相关,下调42个,差异表达蛋白多集中在红细胞吸收氧气并释放二氧化碳、超分子纤维组织、表皮细胞分化、核心组织、NABA核心母体、对无机物的反应、Nop56p相关前rRNA复合体、中性粒细胞迁移、DNA结合的调控、水解酶活性负调节、脂肪酸衍生物代谢过程、上皮细胞增殖的正调控和生物氧化等通路。结论:血瘀证肿瘤组织与正常组织间的差异表达蛋白,其中红细胞吸收氧气并释放二氧化碳、角化、核心组织、超分子纤维组织、血压调节和整合素途径等生物过程相关蛋白可能对血瘀证的形成产生重要作用。
English Summary:
      To compare the differentially expressed proteins between tumor tissues and normal tissues in colorectal cancer with blood stasis syndrome.Methods:The syndromes of colorectal cancer patients were analyzed by clinical investigation method.Postoperative tumor tissues and distal normal intestinal mucosa tissues of patients with blood stasis syndrome were collected.The differentially expressed proteins in the two tissues were compared with iTRAQ quantitative proteomics,and Metascape was used for enrichment and analysis.Results:The comparison of tissues in blood stasis syndrome revealed that there were 55 differential expressed proteins in the distal normal tissues,including 13 up-regulated ones whose functions were related to regulatory exocytosis and 42 down-regulated ones.Differentially expressed proteins were concentrated in erythrocyte uptake of oxygen and release of carbon dioxide,supramolecular fiber tissues,epidermal cell differentiation,nuclear tissues,NABA core matrisome,reactions to inorganic matter,Nop56p-associated pre-rRNA complex,neutrophil migration,regulation of DNA binding,negative regulation of hydrolase activity,metabolic processes of fatty acid derivatives,positive regulation of epithelial cell proliferation,biological oxidation,and other pathways.Conclusion:In the differentially expressed proteins in tumor tissues and normal tissues with blood stasis syndrome,those related to the biological processes of erythrocyte uptake of oxygen and release of carbon dioxide,cornification,nuclear tissues,supramolecular fiber tissues,blood pressure regulation,and integrin pathway may play an important role in the formation of blood stasis syndrome.
