目的:基于“方证对应”理论探讨益气养阴方对气阴两虚高血糖大鼠的功效。方法:选取雄性SD大鼠48只,根据基础血糖水平分为空白组、模型组、十味消渴胶囊阳性药组、益气养阴方低、中、高剂量组,每组8只。采用甲状腺素皮下注射、连续高脂饲料喂养合并链脲佐菌素腹腔注射复合因素造模。实验第一天开始给药,连续灌胃30 d。观察大鼠一般情况,测量体质量,检测糖脂代谢相关指标及气阴两虚证相关生物学指标。结果:与空白组比较,模型组大鼠出现“三多一少”症状及气阴两虚证候表现;空腹血糖(FBG)、血糖曲线下面积(AUC)、胰岛素抵抗指数(HOMA-IR)、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-Ch)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、环磷酸腺苷(cAMP)、cAMP/环磷酸鸟苷(cGMP)显著升高(P<0.05,P<0.01);体质量、胰岛B细胞功能指数(HOMA-β)、胰岛素敏感指数(ISI)、糖化血红蛋白(HbA1c)、空腹胰岛素(FINS)、高密度脂蛋白胆固醇(HDL-Ch)、cGMP、脾脏指数、胸腺指数、抓力指数、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、胃动素(MTL)、促胃液素(GAS)显著降低(P<0.05,P<0.01)。与模型组比较,益气养阴方低、中、高剂量组大鼠气阴两虚证候改善;体质量、脾脏指数、胸腺指数、FINS、HOMA-β、ISI、cGMP、MTL、GAS、IgG、IgM显著升高(P<0.05,P<0.01);FBG、AUC、HOMA-IR、T3、T4、cAMP、cAMP/cGMP显著降低(P<0.05,P<0.01);益气养阴方高剂量组抓力指数显著升高(P<0.01,P<0.05);益气养阴方低剂量组TG显著降低,HDL-Ch显著升高(P<0.05);益气养阴方高剂量组TG、TC、LDL-Ch显著降低(P<0.05)。结论:益气养阴方具有改善气阴两虚型高血糖大鼠证候及糖脂代谢紊乱的功效。 |

To explore the efficacy of Yiqi Yangyin Prescription(YQYY) on hyperglycemic rats with qi and yin deficiency based on prescription-syndrome correspondence.Methods:A total of 48 male SD rats were divided into blank group,model group,positive drug(Shiwei Xiaoke Capsule) group,and YQYY low-,medium-and high-dose groups according to basal blood glucose,with 8 in each group.The hyperglycemic rat model of insulin resistance was established by thyroxine(sc)+continuous high-fat diet feeding+streptozotocin(STZ,ip).The drugs were administered for 30 consecutive days from the first day of the experiment.The general condition of the rats was observed,and the body weight was measured.The related indexes of glucose and lipid metabolism and the biological indexes related to qi and yin deficiency were detected.Results:Compared with the blank group,the model group presented qi and yin deficiency with increased thirst,urination and hunger as well as weight loss.And fasting blood glucose(FBG),area under the curve(AUC),homeostatic model assessment of insulin resistance(HOMA-IR),total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-Ch),triiodothyronine(T3),thyroxine(T4),cyclic adenosine monophosphate(cAMP) and cAMP/cyclic guanosine monophosphate(cGMP) in the model group were higher(P<0.05,P<0.01),while body weight,HOMA-β,insulin sensitivity index(ISI),glycosylated hemoglobin(HbA1c),fasting insulin(FINS),high-density lipoprotein cholesterol(HDL-Ch),cGMP,spleen index,thymus index,grip index,immunoglobulin G(IgG),(IgM),motilin(MTL) and gastrin(GAS) were lower(P<0.05,P<0.01) than the blank group.Compared with the conditions in the model group,qi and yin deficiency was alleviated in the YQYY groups,and body weight,spleen index,thymus index,FINS,HOMA-β,ISI,cGMP,MTL,GAS,IgG and IgM were increased(P<0.05,P<0.01),while FBG,AUC,HOMA-IR,T3,T4,cAMP and cAMP/cGMP were decreased(P<0.05,P<0.01).The YQYY high-dose group had elevated grip index(P<0.01,P<0.05) while reduced TG,TC and LDL-Ch(P<0.05).The YQYY low-dose group had down-regulated TG while up-regulated HDL-Ch(P<0.05).Conclusion:YQYY relieves the syndromes of hyperglycemic rats with qi and yin deficiency and regulates the glucose and lipid metabolism disorders. |
