目的:通过建立气阴两虚证小鼠模型,探讨铁皮枫斗颗粒的益气养阴功效与作用机制。方法:将60只雄性ICR小鼠随机分为空白组、模型组、玉屏风颗粒组、铁皮枫斗颗粒高、中、低剂量组,每组10只,连续给药30 d。实验采用单日限制饮食,双日负重力竭游泳运动,结合灌胃左甲状腺素钠片构建小鼠气阴两虚证模型。动态观察小鼠一般状态,测定小鼠体质量、脏器指数、四肢抓力和力竭游泳时间;采用酶联免疫吸附试验(ELISA)测定小鼠血清环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、白细胞介素-6(IL-6)、补体成分3(C3)、皮质醇(CORT)、促肾上腺皮质激素(ACTH)、促肾上腺皮质激素释放激素(CRH)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、促甲状腺激素(TSH)、雌二醇(E2)和睾酮(T)相关指标,采用实时聚合酶链反应(Real-time PCR)观察下丘脑组织中蛋白激酶A(PKA)和cAMP反应元件结合蛋白(CREB)、信使核糖核酸(mRNA)表达水平。结果:与模型组比较,各给药组均可改善小鼠的一般状态,实验第30天,铁皮枫斗颗粒高、中剂量组均可增加小鼠体质量(P<0.01)。铁皮枫斗颗粒高剂量组可以增大小鼠胸腺指数、脾脏指数和四肢抓力,延长负重游泳力竭时间(P<0.05,P<0.01),降低血清cAMP、cAMP/cGMP、T3、E2、E2/T和IL-6水平(P<0.05,P<0.01),升高血清T、C3水平(P<0.01),降低下丘脑组织PKAmRNA表达水平(P<0.01)。铁皮枫斗颗粒中剂量组可以升高血清中TSH水平(P<0.01),低剂量组可以降低下丘脑组织中CREBmRNA的表达水平(P<0.05)。结论:铁皮枫斗颗粒可以改善气阴两虚证小鼠的一般状态,通过调节环核苷酸、内分泌和免疫系统这一网络机制,上调PKA/CREB信号通路,起到益气养阴的功效。 |

To explore the effect and mechanism of Tiepi Fengdou Granules(TPFD) for replenishing qi and nourishing yin by establishing a mouse model of qi and yin deficiency.Methods:A total of 60 male ICR mice were randomly divided into blank group,model group,Yupingfeng Granules group,and TPFD high-,medium-and low-dose groups,with 10 in each group,and were administered for 30 consecutive days.In the experiment,a mouse model of qi and yin deficiency was established by providing the mice time restricted eating on odd days and weight-bearing exhaustive swimming on even days in combination with levothyroxine sodium tablets(ig).The general state of the mice was dynamically observed,and the body weight,organ index,limb grip strength and exhaustive swimming time were determined.The related indexes of cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP),interleukin-6(IL-6),complement component 3(C3),cortisol(CORT),adrenocorticotropic hormone(ACTH),corticotropin releasing hormone(CRH),triiodothyronine(T3),thyroxine(T4),thyroid-stimulating hormone(TSH),estradiol(E2) and testosterone(T) in serum were detected by enzyme linked immunosorbent assay(ELISA).The mRNA expressions of protein kinase A(PKA) and cyclic-AMP response element binding protein(CREB) in hypothalamus were detected by real-time quantitative polymerase chain reaction(Real-time PCR).Results:Compared with the model group,the administration groups had improved general state.On the 30th day of the experiment,the body weight in both TPFD high-and medium-dose groups was increased(P<0.01).High-dose TPFD increased the thymus index,spleen index and limb grip strength of the mice,prolonged the exhaustive swimming time(P<0.05,P<0.01),reduced the serum levels of cAMP,cAMP/cGMP,T3,E2,E2/T and IL-6(P<0.05,P<0.01) and the mRNA expression of PKA in hypothalamus(P<0.01),and increased the serum levels of T and C3(P<0.01).TPFD up-regulated the serum level of TSH at a medium dose(P<0.01) while down-regulated the mRNA expression of CREB in hypothalamus at a low dose(P<0.05).Conclusion:TPFD improves the general state of mice with qi and yin deficiency.It up-regulates the PKA/CREB signaling pathway by regulating the cyclic nucleotide-endocrine system-immune system network,to exert the effect of replenishing qi and nourishing yin. |
