    目的:采用网络药理学方法探索薏苡附子败酱散治疗盆腔炎性疾病(PID)潜在作用机制,并通过动物实验对相关炎症指标进行验证。方法:通过“中药系统药理学数据库与分析平台(TCMSP)”检索薏苡附子败酱散成分和靶点,人类基因数据库(GeneCard)及在线人类孟德尔遗传数据库(OMIM)检索PID靶基因。可视化工具绘制“疾病-药物”蛋白互作用网络图,行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析,通过动物实验对相关炎症指标进行验证。结果:薏苡附子败酱散治疗PID有21个有效成分及197个靶基因,“疾病-药物”有47个交集基因。GO富集有77个条目,主要与氧化应激反应、上皮细胞增殖等有关,KEGG富集分析有125条通路,主要涉及糖基化终产物受体配体通路、液体剪切应力和动脉粥样硬化、促分裂素原活化蛋白激酶信号通路等。这些通路与炎症反应相关。故由此推测薏苡附子败酱散可能通过抗炎治疗PID。动物实验证明:薏苡附子败酱散可降低PID大鼠血清及子宫组织中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及提高白细胞介素-10(IL-10)水平及表达,改善结果呈剂量依赖性关系。结论:薏苡附子败酱散可通过调节IL-6、IL-10、TNF-α水平及其蛋白表达情况而发挥治疗PID的作用。

    Abstract:

    To explore the potential mechanism of Yiyi Fuzi Baijiang San in the treatment of pelvic inflammatory diseases(PID) by network pharmacology,and to verify the related inflammatory indexes by animal experiments.Methods:The components and targets of Yiyi Fuzi Baijiang San were searched by “Traditional Chinese Medicine System Pharmacology Database and Analysis platform(TCMSP)”.The target genes of PID were searched by Genecard and OMIM database.The “disease-drug” protein-protein interaction network map was drawn by visualization tools and enriched and analyzed by Gene Ontology(GO) functional enrichment analysis and Kyoto Encyclopedia of Gene and Genome(KEGG) pathway enrichment analysis.The relevant inflammatory indexes were verified by animal experiments.Results:There were 21 active components and 197 target genes in the treatment of PID with Yiyi Fuzi Baijiang San and 47 overlapping genes in the “disease-drug” network.There were 77 items in GO enrichment,which were mainly related to oxidative stress and epithelial cell proliferation.KEGG enrichment analysis had 125 pathways,including advanced glycation end product receptor-ligand pathway,liquid shear stress and atherosclerosis,mitogen-activated protein kinase signal pathway,and so on.These pathways were associated with inflammation.Therefore,it was speculated that Yiyi Fuzi Baijiang San may treat PID with anti-inflammation.Animal experiments showed that Yiyi Fuzi Baijiang San decreased the levels of interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in serum and uterine tissues of rats with PID and increased the level and expression of interleukin-10(IL-10) in a dose-dependent manner.Conclusion:Yiyi Fuzi Baijiang San can treat PID by regulating the levels of IL-6,IL-10,TNF-α,and protein expression.
