目的:研究通脉化浊汤对冠心病模型大鼠白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、血管内皮生长因子(VEGF)、细胞间黏附分子-1(ICAM-1)表达的影响。方法:取100只清洁级SD大鼠以随机数字表法分成对照组、模型组、低剂量组、中剂量组及高剂量组,每组20只。除对照组外,其余组大鼠均通过冠状动脉结扎法建立冠心病大鼠模型。造模成功后,分别予以4周的相应干预。比较各组大鼠心功能指标水平,血清IL-6、IL-10水平,心肌组织VEGF、ICAM-1表达水平,血脂指标水平。结果:模型组,低剂量组、中剂量组、高剂量组左心室收缩压(LVSP)、左心室收缩期血压最大升高率(+dp/dtmax)、左心室收缩期血压最大下降率(-dop/dtmax)、IL-10、VEGF、高密度脂蛋白水平均低于对照组,且低剂量组、中剂量组、高剂量组上述指标水平高于模型组,中剂量组、高剂量组又高于低剂量组,高剂量组又高于中剂量组(均P<0.05)。模型组、低剂量组、中剂量组、高剂量组IL-6、ICAM-1、胆固醇、低密度脂蛋白水平均高于对照组,且低剂量组、中剂量组、高剂量组低于模型组,中剂量组、高剂量组低于低剂量组,高剂量组低于中剂量组(均P<0.05)。结论:通脉化浊汤可有效下调IL-6、ICAM-1表达水平,同时上调IL-10、VEGF表达水平,这可能是其治疗冠心病的重要机制。 |

To study the effect of Tongmai Huazhuo Decoction(THD) on the expression of interleukin-6(IL-6),interleukin-10(IL-10),vascular endothelial growth factor(VEGF),and intercellular adhesion molecule-1(ICAM-1) in rats with coronary heart disease.Methods:A total of 100 SD rats were randomized into control group,model group,and low-dose,medium-dose,and high-dose THD groups by random number table method,with 20 rats in each group.Coronary artery ligation was performed on rats expect the control group to induce coronary heart disease.After modeling,administration began and lasted 4 weeks.The levels of cardiac function indexes,serum IL-6 and IL-10,expression of VEGF and ICAM-1 in myocardium,and serum lipid level were compared among the groups.Results:The left ventricular systolic pressure(LVSP),maximal rate of rise of left ventricular pressure(+dp/dtmax),maximal rate of decline of left ventricular pressure(-dop/dtmax),and levels of IL-10,VEGF,and high-density lipoprotein were lower in the model group and three THD groups than in the control group,and higher in the three THD groups than in the model group.Moreover,the above indexes were higher in the medium-dose and high-dose THD groups than in the low-dose group and higher in the high-dose group than in the medium-dose group(P<0.05).The levels of IL-6,ICAM-1,cholesterol and low-density lipoprotein in the model group and three THD groups were higher than those in the control group,and the levels in the three THD groups were lower than those in the model group.In addition,the levels were lower in the in the medium-dose and high-dose THD groups than in the low-dose group,and lower in the high-dose group than in the medium-dose group(P<0.05).Conclusion:THD can down-regulate the levels of IL-6 and ICAM-1 and up-regulate the levels of IL-10 and VEGF,which explains the efficacy on coronary heart disease. |
