目的:研究化痰祛瘀方对心房颤动大鼠血清基质金属蛋白酶-2(MMP-2)、金属蛋白酶组织抑制物-2(TIMP-2)含量的影响,探讨化痰祛瘀方防治心房颤动机制。方法:将60只雄性斯泼累格·多雷(SD)大鼠随机分对照组、模型组、化痰祛瘀方高剂量组、中剂量组、低剂量组和维拉帕米组,共6组,造模动物连续7 d采用尾静脉注射氯化乙酰胆碱ACh(66 μg/mL)-CaCl2(10 mg/mL)混合液方式建立心房颤动大鼠模型。2周后使用PowerLab 15T数据采集分析系统记录肢体Ⅱ导联心电图,监测各组心房颤动大鼠心房颤动持续时间;酶联免疫吸附试验法检测大鼠血清中MMP-2、TIMP-2含量。结果:心房颤动持续时间比较:与对照组比较,模型组大鼠心房颤动持续时间明显延长(P<0.05);与模型组比较,各治疗大鼠心房颤动持续时间较模型组明显缩短(P<0.05)。血清中MMP-2、TIMP-2含量比较:与对照组比较,模型组大鼠血清中MMP-2含量升高(P<0.05)、TIMP-2含量降低(P<0.01)。与模型组比较,化痰祛瘀方高剂量组、中剂量组,维拉帕米组大鼠血清中MMP-2含量降低(P<0.05)、TIMP-2含量升高(P<0.05)。结论:化痰祛瘀方可缩短由氯化乙酰胆碱诱导心房颤动大鼠心房颤动持续时间,改变大鼠心肌超微结构变化,降低大鼠血清中MMP-2含量、升高TIMP-2含量,抑制心房颤动所导致心肌纤维化,从而为化痰祛瘀方治疗心房颤动提供了科学的理论依据。 |

To investigate the effect of Huatan Quyu Formula on the serum matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloprotease-2(TIMP-2) levels in rats with atrial fibrillation and explore the mechanism of Huatan Quyu Formula for preventing and treating atrial fibrillation.Methods:Sixty male Sprague-Dawley(SD) rats were randomly divided into a control group,a model group,high-,medium-,and low-dose Huatan Quyu Formula group,and a verapamil group.The atrial fibrillation model was induced by tail vein injection of acetylcholine chloride(ACh,66 μg/mL)-CaCl2(10 mg/mL) for seven days.After two weeks,the duration of atrial fibrillation was monitored using limb lead Ⅱ ECG recorded by the PowerLab 15T data acquisition and analysis system.The serum MMP-2 and TIMP-2 levels were measured by enzyme-linked immunosorbent assay(ELISA).Results:Duration of atrial fibrillation.The duration of atrial fibrillation in the model group was significantly prolonged compared with that in the control group(P<0.05).The duration of atrial fibrillation was significantly shorter in the groups with drug intervention than that in the model group(P<0.05).Serum MMP-2 and TIMP-2 levels.The serum MMP-2 level increased(P<0.05) and the TIMP-2 level decreased(P<0.01) in the model group compared with that in the control group.The serum MMP-2 level decreased(P<0.05) and the TIMP-2 level increased(P<0.05) in the verapamil group,high-and medium-dose Huatan Quyu Formula groups compared with that in the model group.Conclusion:The Huatan Quyu Formula can shorten the duration of atrial fibrillation in ACh-induced rats,change the ultrastructure of rat myocardium,reduce the serum MMP-2 level,increase the TIMP-2 level,and inhibit myocardial fibrosis caused by atrial fibrillation.These results provide a scientific theoretical basis for the treatment of atrial fibrillation with the Huatan Quyu Formula. |
