      目的:观察推拿对运动性疲劳(EF)大鼠外周血炎症介质和外周血单个核细胞(PBMCs)凋亡与亚型平衡的影响,探索推拿改善运动性免疫炎性损伤的机制。方法:选取Wistar大鼠30只,随机将6只作为空白组,其余运用跑台运动法进行造模,将16只成模大鼠随机分为模型组及推拿组,每组8只。空白组不进行任何处理,模型组每日力竭运动后于笼中休息,推拿组每日力竭运动后予以推拿干预1次,共干预21 d。取材后,运用酶联免疫吸附试验(ELISA)法测定外周血白细胞介素-1β(IL-1β)与肿瘤坏死因子-α(TNF-α)含量,流式细胞术测定PBMCs凋亡情况及Th1/Th2,Th17/Treg亚型分布情况。结果:与空白组比较,模型组大鼠外周血中IL-1β和TNF-α的含量显著升高(均P<0.01);与模型组比较,推拿组外周血中IL-1β和TNF-α含量显著下调(均P<0.01)。与空白组比较,模型组大鼠PBMCs凋亡率显著上升(P<0.01);与模型组比较,推拿组大鼠PBMCs凋亡率显著下降(P<0.05)。与空白组比较,模型组大鼠PBMCs中Th1和Th17细胞增多,Th2和调节性T(Treg)细胞减少,出现Th1和Th17漂移,Th1/Th2及Th17/Treg细胞比例均显著上调(均P<0.01);与模型组比较,推拿组大鼠PBMCs中Th1/Th2及Th17/Treg细胞比例均显著下调(P<0.01,P<0.05)。结论:推拿改善了EF大鼠外周血的免疫炎性状态,体现出抗炎、抗凋亡及免疫调节的手法效应。
English Summary:
      To observe the effects of Tuina on peripheral blood inflammatory mediators and peripheral blood mononuclear cell(PBMC) apoptosis and subtype balance in exercise-induced fatigue(EF) rats and explore the mechanism of Tuina in improving exercise-induced immune-inflammatory injury.Methods:Thirty Wistar rats were selected and six rats were assigned to the blank group.The remaining rats underwent model induction using a treadmill exercise method.Sixteen model rats were divided into a model group(n=8) and a Tuina group(n=8).The blank group did not undergo any treatment.The model group rested in the cage after exhaustive exercise every day,and the Tuina group received Tuina intervention once a day after exhaustive exercise for a total of 21 days.Enzyme-linked immunosorbent assay(ELISA) was used to measure the content of peripheral blood interleukin(IL)-1β and tumor necrosis factor(TNF)-α,while flow cytometry was used to measure PBMC apoptosis and Th1/Th2,Th17/Treg subtype distribution.Results:The content of IL-1β and TNF-α in the peripheral blood of the model group significantly increased as compared with that in the blank group(P<0.01).The content of IL-1β and TNF-α in the peripheral blood of the Tuina group significantly decreased as compared with that in the model group(P<0.01).The PBMC apoptosis rate in the model group was significantly higher than that in the blank group(P<0.01),while the PBMC apoptosis rate in the Tuina group was significantly lower than that in the model group(P<0.05).Compared with the blank group,the model group showed increased Th1 and Th17 cells in PBMCs,decreased Th2 and regulatory T(Treg) cells,and shifted Th1 and Th17 cells,resulting in Th1/Th2 and Th17/Treg cell ratio up-regulation(P<0.01).Compared with the model group,the Th1/Th2 and Th17/Treg cell ratios in PBMCs of the Tuina group were significantly down-regulated(P<0.01,P<0.05).Conclusion:Tuina can improve the immunoinflammatory status of peripheral blood in EF rats,showing the effects of anti-inflammatory,anti-apoptosis,and immune regulation.
