      目的:研究葛根汤颗粒对甲型H1N1流感病毒PR8株感染致小鼠病毒性肺炎模型的治疗及死亡保护作用。方法:采用甲型H1N1流感病毒(PR8株)滴鼻感染小鼠造成病毒性肺炎模型,观察小鼠的肺指数和肺指数抑制率,肺内细支气管及肺组织苏木精-伊红(HE)染色病理切片镜下病变观察及等级评分;同时采用流感病毒PR8毒株滴鼻感染小鼠造成死亡保护模型,观察小鼠感染后2周内的死亡情况,计算小鼠的死亡率、死亡保护率、平均存活天数和生命延长率。结果:葛根汤颗粒高、中剂量组可显著降低模型组小鼠肺指数(P<0.01);葛根汤颗粒高、中剂量组与模型组比较可显著减轻支气管炎症反应渗出状况,红肿相对消退,并且缓解病毒感染所致肺泡、肺间质渗出炎症反应及红细胞渗出情况(P<0.01,P<0.05);模型组小鼠死亡率90%,平均存活天数9.95 d,葛根汤颗粒3个剂量组与模型组比较可显著降低小鼠死亡率,延长小鼠平均存活天数,提高生命延长率(P<0.01,P<0.05)。结论:本研究发现葛根汤颗粒可显著降低病毒性肺炎模型小鼠的肺指数,并显著减轻病毒感染所致的小鼠肺内细支气管及肺局部炎症反应损伤,对模型小鼠的肺部炎症反应有明显的治疗作用;同时可显著降低模型小鼠死亡率,延长平均存活天数,提高生命延长率,对感染小鼠有死亡保护作用。
English Summary:
      To study the therapeutic and death protective effects of Gegentang Granules on the mouse model of viral pneumonia caused by infection with influenza A(H1N1) virus PR8 strain.Methods:The mouse model of viral pneumonia was established by nasal dripping with influenza A H1N1 virus(PR8 strain).The lung index and lung index inhibition rate were recorded,and the pathological changes of the bronchus and lung tissue were observed and graded by hematoxylin-eosin staining.At the same time,we established a mouse model of influenza virus PR8 strain infection by nasal dripping to observe the death of mice within 2 weeks after infection,and recorded the mortality rate,death protection rate,average survival days,and life extension rate.Results:Medium- and high-dose Gegentang Granules reduced the lung index(P<0.01) and alleviated bronchial inflammation and exudation,redness and swelling,alveolar and pulmonary interstitial inflammation,and erythrocyte extravasation caused by viral infection(P<0.01,P<0.05) compared with the model group.The mice in the model group showed a mortality rate of 90% and the average survival of 9.95 days.Compared with the model group,the three doses of Gegentang Granules reduced the mortality rate of mice,prolonged the average survival days,and improved the life extension rate of mice(P<0.01,P<0.05).Conclusion:Gegentang Granules can significantly reduce the lung index and alleviate the bronchial and local pulmonary inflammation caused by viral infection,demonstrating an obvious therapeutic effect on the pulmonary inflammation of the model mice.Moreover,it can significantly reduce the mortality,prolong the average survival days,and improve the life extension rate,protecting the infected mice from death.
