      目的:探讨柴归汤对高盐诱导的高血压小鼠的血压、血浆、肾组织血管紧张素转换酶2(ACE2)和肠道菌群的干预作用。方法:本文通过高盐饲料诱导建立小鼠高血压模型,采用贝那普利和柴归汤进行平行对照干预,并检测小鼠血压、血浆ACE2水平,肾脏组织肾素、ACE2水平,以及肠道菌群丰度的变化。结果:1)柴归汤可持续稳定降低高盐诱导的高血压小鼠模型的收缩压和舒张压水平(P<0.001),降压效果与贝那普利相当(P>0.05);2)柴归汤升高了高血压小鼠降低的血浆ACE2水平(P<0.001),但贝那普利组血浆ACE2水平高于柴归汤组(P<0.001);3)柴归汤可降低高血压小鼠的肾脏组织肾素水平和肾素/ACE2比值(P<0.05);4)高血压小鼠肠道Bacteroidia菌群丰度降低,而Clostridia菌群丰度升高。贝那普利和柴归汤可以改善菌群失衡(P<0.05)。结论:柴归汤对高盐诱导的高血压小鼠具有良好的降血压作用,其机制与柴归汤升高血浆ACE2水平、降低肾脏组织肾素水平和肾素/ACE2比值、调节肠道菌群有关。
English Summary:
      To investigate the intervention effect of Chaigui Decoction on blood pressure,levels of angiotensin-converting enzyme 2(ACE2) in plasma and the renal tissue,and intestinal flora in high-salt-induced hypertensive mice.Methods:In this study,a high-salt diet was used to induce a hypertension model in mice.Benazepril and Chaigui Decoction were administered in parallel for intervention,and the blood pressure,plasma ACE2 levels,renin and ACE2 levels in the renal tissue,and changes in intestinal flora abundance were measured.Results:1) Chaigui Decoction could continuously and stably reduce systolic and diastolic blood pressure levels in the high-salt-induced hypertensive mouse model(P<0.001),with an antihypertensive effect comparable to that of benazepril(P>0.05). 2) Chaigui Decoction increased the decreased plasma ACE2 levels in hypertensive mice(P<0.001),but the plasma ACE2 levels in the benazepril group were higher than those in the Chaigui Decoction group(P<0.001). 3) Chaigui Decoction reduced renal tissue renin levels and the renin/ACE2 ratio in hypertensive mice(P<0.05). 4) The abundance of Bacteroidia intestinal flora decreased,while Clostridia increased in hypertensive mice.Both benazepril and Chaigui Decoction improved the imbalance of intestinal flora(P<0.05).Conclusion:Chaigui Decoction has a good antihypertensive effect on high-salt-induced hypertensive mice,and its mechanism is related to the elevation of plasma ACE2 levels,reduction of renal tissue renin levels and renin/ACE2 ratio,and regulation of intestinal flora.
