To observe the effect of Bushen Shengxue Formula on cisplatin-induced damage to bone marrow mesenchymal stem cells(BMSCs) and explore the mechanism of Bushen Shengxue Formula in improving bone marrow suppression after chemotherapy.Methods:Fifty male SD rats were randomly divided into a blank control group and a Bushen Shengxue Formula group,with 25 rats in each group.Bushen Shengxue Formula-medicated serum and blank serum were prepared,and primary cultures of BALB/c mouse BMSCs were established.BMSCs were randomly divided into groups,and the intervention concentrations of cisplatin and Bushen Shengxue Formula-medicated serum were determined using the CCK-8 method.After 48 hours of intervention,flow cytometry was used to detect cell apoptosis rate and cell cycle,and the enzyme-linked immunosorbent assay was used to measure the levels of hematopoietic regulatory factors in the culture medium.Results:After 48 hours of intervention,the apoptosis rate of bone marrow cells increased significantly(P<0.01).In the Bushen Shengxue Formula-medicated serum group,the proportion of cells in the G1 phase decreased significantly(P<0.05),the proportion of cells in the S phase increased significantly(P<0.05),and the proportion of proliferating cells increased significantly(P<0.05).The Bushen Shengxue Formula-medicated serum group showed a significant decrease in the apoptosis rate of bone marrow cells(P<0.001),a significant decrease in the proportion of cells in the G1 phase(P<0.05),a significant increase in the proportion of proliferating cells(P<0.05),and a significant increase in IL-3 level(P<0.01),and decreased levels of TNF-α,TGF-β,and IFN-γ(all P<0.05).Conclusion:Bushen Shengxue Formula can alleviate cisplatin-induced apoptosis of BMSCs,promote cell proliferation,stimulate the secretion of hematopoietic growth factors,and reduce the secretion of hematopoietic inhibitory factors.These results suggest that this formula has a certain improving effect on bone marrow suppression after chemotherapy.
