To investigate the effect of Baohe Xiaoji Formula on the proliferation and migration of gastric cancer SGC-7901 cells and elucidate its molecular mechanism at the mRNA and protein levels.Methods:Gastric cancer SGC-7901 cells in the logarithmic growth phase were selected and divided into a blank control group and Baohe Xiaoji Formula groups at different concentrations(0.5,1,2,4,8,12,and 16 mg/mL).The MTT assay was used to detect cell proliferation at different time points(24,48,and 72 hours).The Transwell assay was performed to assess the migration ability of SGC-7901 cells.Flow cytometry was used to evaluate cell apoptosis.Quantitative PCR and Western blot were conducted to measure the mRNA and protein expression levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(Akt),mammalian target of rapamycin(mTOR),and vascular endothelial growth factor(VEGF) related to cell proliferation,migration,and angiogenesis in gastric cancer SGC-7901 cells.Results:Compared with the blank control group,the Baohe Xiaoji Formula groups showed increased proliferation inhibition rates of gastric cancer SGC-7901 cells after different durations of treatment(all P<0.05),reduced number of transmembrane cells(all P<0.05),inhibited tubule formation(all P<0.05),and decreased mRNA and protein expression levels of Akt,mTOR,PI3K,and VEGF(all P<0.05).Conclusion:Baohe Xiaoji Formula inhibits the proliferation and migration of SGC-7901 gastric cancer cells and promotes cell apoptosis by suppressing the mRNA and protein expression of Akt,mTOR,PI3K,and VEGF.
