Based on the phosphatidylinositol3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR) signaling pathway,the effect of acupuncture on apoptosis of ectopic endometrial histiocytes in rats with endometriosis(EMs) and related mechanisms were explored.Methods:The animal model of EMs was constructed by autologous transplantation,which was divided into the model group,control group,and acupuncture group according to the random number table method,with 12 animals in each grou.The acupuncture group was given acupuncture once a day for 28 days,and no treatment regimen was given to the control group and the model group.The histopathological morphology of the intima of rats was observed by hematoxylin-eosin(HE) staining.TdT-mediated dUTP nick-end labeling(TUNEL) was used to observe apoptosis changes in in-situ and ectopic endometrial histiocytes.Real-time polymerase chain reaction(RT-PCR) and western blot were used to detect gene expression and protein content changes in estrogen receptor α(ER-α),estrogen receptor β(ER-β),PI3K,AKT,mTOR,cysteine aspartate protease-3(Caspase-3),Bcl-2-associated X protein(Bax),and B lymphocyte tumor 2(Bcl-2) in in-situ and ectopic endometrial tissue.Results:Compared with the control group,the model group had significant ectopic hyperplasia and inflammatory factor infiltration,and the expression and protein content of PI3K,AKT,and mTOR genes were significantly increased(P<0.05).After the treatment intervention,compared with the model group,cell proliferation and inflammatory factor infiltration were significantly alleviated,while the gene expression and protein content of PI3K,AKT,mTOR,and Bcl-2 were significantly reduced,and those of Caspase-3 and Bax were significantly increased(P<0.05).Conclusion:Acupuncture can accelerate apoptosis in ectopic lesion tissue,which may be closely related to the regulation of the PI3K/AKT/mTOR signaling pathway to promote related apoptotic factors.
