目的:探讨和胃安神方对失眠模型大鼠下丘脑5-羟色胺(5-HT)、多巴胺(DA)和去甲肾上腺素(NE)水平的影响。方法:将60只雄性Wistar大鼠随机分为6组,即正常对照组、失眠模型组、地西泮组、和胃安神方低剂量组、和胃安神方中剂量组、和胃安神方高剂量组。采用酶联免疫吸附试验(ELISA)检测各组大鼠下丘脑组织5-HT、DA、NE的水平。结果:与正常对照组比较,失眠模型组大鼠5-HT水平显著下降,差异有统计学意义(P<0.05),DA、NE水平较正常对照组显著升高,差异有统计学意义(P<0.05);与失眠模型组比较,地西泮组、和胃安神方低剂量组5-HT、DA、NE水平差异均无统计学意义(均P>0.05);和胃安神方中剂量组、和胃安神方高剂量组5-HT水平均较失眠模型组显著升高,差异有统计学意义(P<0.05),DA、NE水平较失眠模型组显著下降,差异有统计学意义(P<0.05)。结论:和胃安神方治疗失眠症的疗效机制可能与调节脑内5-HT、DA、NE水平有关。 |

To investigate the effects of Hewei Anshen Formula(HWAS) on the levels of 5-hydroxytryptamine(5-HT),dopamine(DA),and norepinephrine(NE) in the hypothalamus of insomnia model rats.Methods:Sixty male Wistar rats were randomly divided into six groups:a normal control group,an insomnia model group,a diazepam group,and low-,medium-,and high-dose HWAS groups.Enzyme-linked immunosorbent assay(ELISA) was used to detect the levels of 5-HT,DA,and NE in the hypothalamic tissues of the rats in each group.Results:The 5-HT level in the hypothalamus of rats in the insomnia model group was significantly decreased(P<0.05),while the levels of DA and NE were significantly increased(P<0.05) as compared with those in the normal control group.There were no significant differences in 5-HT,DA,and NE levels in the diazepam group and the low-dose HWAS group when compared with the insomnia model group(P>0.05).The medium-dose and high-dose HWAS groups showed significantly higher 5-HT levels than the insomnia model group(P<0.05),while the levels of DA and NE were significantly decreased(P<0.05).Conclusion:The therapeutic mechanism of the HWAS for treating insomnia may be related to the regulation of 5-HT,DA,and NE levels in the brain. |
