      目的:探讨隔药饼灸调控子宫内膜异位症(EMT)小鼠对子宫内膜磷脂酰肌醇3激酶(PI3K)信号通路表达的影响。方法:将42只6~8周龄雌性重度联合免疫缺陷病(SCID)小鼠按照随机区组分组法分为正常组(n=8)和造模组(n=34)。造模组小鼠均采用EMT自体移植法进行造模,按照随机数字表法选取2只小鼠验证造模成功后,按随机区组分组法分成EMT模型组、隔药饼灸组、西药组和艾灸加西药组4组,每组8只,并给予相应治疗。观察评估各组小鼠扭体反应及囊泡抑制率,采用蛋白质印迹法检测小鼠异位内膜组织中PI3K、蛋白激酶B(AKT)、哺乳动物雷帕霉素靶蛋白(mTOR)的表达。结果:与正常组比较,模型组、西药组、隔药饼灸组、艾灸加西药组小鼠的扭体反应次数升高,差异均有统计学意义(均P<0.01),与西药组比较,隔药饼灸组和艾灸加西药组扭体反应次数下降,差异均有统计学意义(均P<0.05);与模型组比较,西药组、隔药饼灸组、艾灸加西药组治疗前后异位子宫组织体积明显缩小,差异均有统计学意义(均P<0.05);与模型组异位内膜组织比较,西药组、隔药饼灸组、艾灸加西药组异位内膜组织中的PI3K、AKT、mTOR表达降低,差异均有统计学意义(均P<0.05);与隔药饼灸组比较,西药组、艾灸加西药组异位内膜组织中的PI3K、AKT、mTOR表达均升高,差异均有统计学意义(均P<0.05)。结论:隔药饼灸可能是通过下调小鼠子宫异位内膜组织PI3K/AKT/mTOR高表达而达到镇痛和抑制囊泡生长作用。
English Summary:
      This study aims to explore the effects of medicinal cake-separated moxibustion on the expression of the phosphatidylinositol-3-kinase(PI3K) signaling pathway in mice with endometriosis(EMT).Methods:A total of 42 female mice with server combined immune deficiency(SCID) aged 6~8 weeks were randomly divided into two groups:normal group(eight mice) and model-making group(34 mice).The EMT autologous transplantation method was adopted to design the model in the model-making group.After model verification by randomly selecting two mice,the mice were randomly divided into four groups,namely EMT model group,medicinal cake-separated moxibustion group,western medicine group,and moxibustion plus western medicine group,with eight mice in each group,and all the mice were given corresponding treatment.The writhing response and vesicle inhibition rate of mice in each group were observed,and Western blot was used to detect the protein expression of PI3K,protein kinase B(AKT),and mammalian target of rapamycin(mTOR) in the ectopic endometrial tissue of mice.Results:Compared with the number of writhes in mice of the normal group,the number of writhes increased in the model group,western medicine group,medicinal cake-separated moxibustion group,and moxibustion plus western medicine group,and the difference was statistically significant(P<0.01).Compared with the number of writhes in mice of the western medicine group,the number of writhes decreased in the medicinal cake-separated moxibustion group and moxibustion plus western medicine group,and the difference was statistically significant(P<0.05).Compared with mice in the model group,the volume of ectopic uterine tissue was significantly reduced after treatment in mice of the western medicine group,medicinal cake-separated moxibustion group,and moxibustion plus western medicine group,and the difference was significant(P<0.05).Compared with the model group,the expression of P3K,AKT,and mTOR decreased in the western medicine group,medicinal cake-separated moxibustion group,and moxibustion plus western medicine group,and the difference was statistically significant(P<0.05).Compared with the moxibustion plus western medicine group,the expression of P3K,AKT,and mTOR decreased in the western medicine group and medicinal cake-separated moxibustion group,and the difference was statistically significant(P<0.05).Conclusion:The medicinal cake-separated moxibustion has an analgesic effect and inhibits the growth of vesicles by reducing the high expression of PI3K/AKT/mTOR in the ectopic endometrial tissue of mice.
