      目的:探索鼻敏宁颗粒对寒证变应性鼻炎大鼠鼻黏膜组织中黏蛋白5B(MUC5B)、黏蛋白5AC(MUC5AC)、Toll样受体2(TLR2)蛋白表达情况。方法:将60只大鼠随机分成空白对照组、模型组、阳性对照组、鼻敏宁低、中、高剂量组。造模成功后,鼻敏宁颗粒高剂量组大鼠予鼻敏宁颗粒2 g/kg,鼻敏宁颗粒中剂量组1 g/kg,鼻敏宁颗粒低剂量组0.5 g/kg,阳性对照组予辛芩颗粒5 g/kg,2 mL/次,空白对照组与模型组给予同等容积的0.9%氯化钠注射液灌胃,均为每天1次,连续7 d。应用免疫组织化学定位检测鼻黏膜上皮细胞TLR2蛋白表达情况,应用蛋白质印迹法(WB)检测鼻黏膜上皮组织MUC5B及MUC5AC蛋白表达量。结果:鼻敏宁低中高剂量组中阳性染色明显减弱,模型组与其他组比较,差异有统计学意义(P<0.05);与模型组比较,中剂量组与高剂量组MUC5B及MUC5AC含量显著降低,有明显的差异有统计学意义(P<0.05)。结论:鼻敏宁颗粒可通过降低黏蛋白5B及黏蛋白5AC的表达,TLR2蛋白的表达,减轻大鼠鼻黏膜免疫损伤,达到治疗变应性鼻炎的作用。
English Summary:
      To explore the expression of mucin 5B(MUC5B),mucin 5AC(MUC5AC),and Toll-like receptor 2(TLR2) in the nasal mucosa of the rats with allergic rhinitis(AR) due to cold syndrome and treated with Biminning Granules.Methods:Sixty rats were randomized into 6 groups:blank control,model,positive control(Xinqin Granules,5 g/kg,2 mL per time),and low-,medium-,and high-dose(0.5,1,and 2 g/kg,respectively) Biminning Granules.The successfully modeled rats were administrated with corresponding drugs or 0.9% sodium chloride(blank control and model groups) by gavage once a day for 7 days.The immunohistochemical assay was employed to localize the expression of TLR2 in the nasal mucosa.Western blotting was employed to determine the expression levels of MUC5B and MUC5AC in the nasal mucosa.Results:Positive staining was significantly attenuated in the Biminning groups,and the model group showed significant differences compared with other groups(P<0.05).Compared with the model group,medium and high-dose Biminning groups showcased down-regulated expression of MUC5B and MUC5AC(P<0.05).Conclusion:Biminning Granules can reduce the immune damage to the nasal mucosa by down-regulating the expression of MUC5B,MUC5AC,and TLR2,thereby treating AR in rats.
