To investigate the role of microRNA-665(miR-665)/cold-inducible RNA-binding protein(CIRBP) in endometrial fibrosis in intrauterine adhesion(IUA) and the regulatory effect of Bushen Huoxue Decoction.Methods:TargetScan was used to analyze the binding of CIRBP with miR-665.Quantitative real-time PCR and Western blotting were employed to determine the transcription and expression levels of CIRBP and miR-665 in cells and animal models.The dual-luciferase reporter system and overexpression vector were utilized to examine the mutual regulation between CIRBP and miR-665,as well as the regulatory effects of Bushen Huoxue Decoction on miR-665/CIRBP.Enzyme-linked immunosorbent assay and hematoxylin-eosin staining were performed to measure the levels of relevant factors and pathological changes in the rat model of IUA.Results:The rat model of IUA showed upregulated transcription level of CIRBP and downregulated transcription level of miR-665.The in vitro experiments showed that miR-665 mimics downregulated the expression level of CIRBP in endometrial stromal cells.Bushen Huoxue Decoction lowered the levels of transforming growth factor-β1(TGF-β1),estradiol(E2),and tumor necrosis factor-α(TNF-α) in the serum of the rat model.Additionally,Bushen Huoxue Decoction upregulated the transcription level of miR-665 while downregulating the expression level of CIRBP,thereby alleviating the pathological changes in the endometrial tissue of the rats.Conclusion:Bushen Huoxue Decoction alleviates endometrial fibrosis in the rat model of IUA by regulating the miR-665/CIRBP pathway,which provides new strategies and directions for the treatment of IUA.
