To investigate the protective effects of oral administration of Xuebijing Injection on intestinal barrier function in septic mice and analyze the potential mechanisms of oral Xuebijing Injection in improving intestinal barrier function.Methods:Male ICR mice were randomly divided into control group(NC),model group(M),oral Xuebijing Injection group(XBJ-ig),and intravenous Xuebijing Injection group(XBJ-iv) according to ramdom block method.The sepsis-induced intestinal barrier dysfunction model was established by injecting lipopolysaccharide(LPS) into the dorsal penile vein.The study assessed survival rate,serum levels of tumor necrosis factor-alpha(TNF-α) and interleukin-1 beta(IL-1β),histopathological changes in the ileum,intestinal permeability,and expression of tight junction proteins Claudin-1 and ZO-1 in the ileum.Results:Compared to the model group,the oral Xuebijing Injection group showed an improvement in the survival rate of septic mice.Serum levels of inflammatory mediators TNF-α and IL-1β were significantly increased in the model group compared to the control group(P<0.01),along with a marked increase in intestinal permeability(P<0.01) and a significant downregulation of Claudin-1 and ZO-1 protein expression(P<0.05).Compared to the model group,the oral Xuebijing Injection group exhibited significantly reduced levels of TNF-α and IL-1β(P<0.05).Six hours after modeling,the intestinal mucosal permeability of the oral Xuebijing Injection group began to show significant improvement(P<0.05),along with a significant upregulation of Claudin-1 and ZO-1 protein expression(P<0.05).Conclusion:Oral administration of Xuebijing Injection improves intestinal barrier function in septic mice by enhancing the expression of tight junction proteins.
