    摘要:

    目的:探讨托里消毒散对高糖诱导损伤的M2型巨噬细胞分泌抗炎因子和损伤修复因子的影响。方法:利用佛波酯(PMA)、白细胞介素-4(IL-4)和白细胞介素13(IL-13)体外诱导人单核细胞THP-1向M2型巨噬细胞分化,通过观察细胞形态、蛋白质免疫印迹法测定M2型巨噬细胞表面标志物精氨酸酶1(Arg-1)、CD206蛋白的表达以鉴定所诱导的细胞。将鉴定成功的M2型巨噬细胞用33 mmol/L葡萄糖干预120 h后随机分为观察组和模型组,观察组用500 μg/mL托里消毒散进行干预,模型组用等体积的磷酸盐缓冲液(PBS)处理;同时设置正常组,用等体积PBS干预。各组干预48 h后收集细胞上清液,采用酶联免疫吸附实验(ELISA)检测白细胞介素-10(IL-10)、IL-4、IL-13、转化生长因子β1(TGF-β1)、内皮生长因子(VEGF)、肝细胞生长因子(HGF)、胰岛素样生长因子-1(IGF-1)、血小板衍生因子(PDGF)及成纤维细胞生长因子(FGF)的吸光度。结果:光镜及蛋白质印迹法结果显示,人单核细胞THP-1成功极化得到M2型巨噬细胞。ELISA结果显示,与正常组相比,模型组IL-10、IL-4、IL-13、VEGF、HGF、IGF-1、TGF-β1、PDGF、FGF含量明显降低,差异有统计学意义(P<0.05);与模型组相比,观察组中IL-10、IL-4、IL-13、VEGF、HGF、IGF-1、PDGF、FGF含量明显升高,差异有统计学意义(P<0.05),TGF-β1差异无统计学意义。结论:托里消毒散能够促进高糖诱导损伤的M2型巨噬细胞分泌抗炎因子和损伤修复因子,可用于糖尿病皮肤溃疡创面的修复。

    Abstract:

    To investigate the effect of Tuoli Xiaodu Powder on the secretion of anti-inflammatory and tissue repair factors in M2 macrophages damaged by high glucose induction.Methods:Human monocytic THP-1 cells were induced to differentiate into M2 macrophages in vitro using phorbol 12-myristate 13-acetate(PMA),interleukin-4(IL-4),and interleukin-13(IL-13).The induced cells were identified by observing their morphology and detecting the expression of M2 macrophage surface markers arginase-1(Arg-1) and CD206 via Western blot.Successfully identified M2 macrophages were exposed to 33 mmol/L glucose for 120 h and then randomly divided into an experimental group and a model group.The experimental group was treated with 500 μg/mL Tuoli Xiaodu Powder,while the model group received an equivalent volume of phosphate-buffered saline(PBS).A normal group was also established,receiving the same volume of PBS as an intervention.After 48 h of intervention,the supernatants of the cell cultures were collected,and enzyme-linked immunosorbent assay(ELISA) was used to measure the absorbance of interleukin-10(IL-10),IL-4,IL-13,transforming growth factor-β1(TGF-β1),vascular endothelial growth factor(VEGF),hepatocyte growth factor(HGF),insulin-like growth factor-1(IGF-1),platelet-derived growth factor(PDGF),and fibroblast growth factor(FGF).Results:Microscopy and Western blot analysis confirmed the successful polarization of THP-1 monocytes into M2 macrophages.ELISA results showed that compared to the normal group,the model group exhibited significantly lower levels of IL-10,IL-4,IL-13,VEGF,HGF,IGF-1,TGF-β1,PDGF,and FGF(P<0.05).Compared to the model group,the experimental group showed significantly increased levels of IL-10,IL-4,IL-13,VEGF,HGF,IGF-1,PDGF,and FGF(P<0.05),while the difference in TGF-β1 levels was not statistically significant.Conclusion:Tuoli Xiaodu Powder promotes the secretion of anti-inflammatory and tissue repair factors in M2 macrophages damaged by high glucose induction,suggesting its potential application in the treatment of diabetic skin ulcer wounds.
